Resumen

El trabajo describe la estructura cristalográfica del complejo ribonucleoproteíco Vault, de ~13 MDa, determinado a 8 Å de resolución junto con la del dominio N-terminal de la proteína estructural MVP que forma el barril central de la partícula, resuelta a 2.1 Å. La combinación de las dos estructuras ha permitido elucidar las interacciones deteminantes para el ensamblado de las dos mitades de la partícula y describir un modelo de apertura desencadenado por un cambio de pH. Aunque las Vaults se relacionan con proliferación celular y resistencia a multitud de fármacos antitumorales, sus funciones específicas continúan siendo un enigma. La comprensión de su estructura es el primer paso para esclarecer las funciones de este abundante complejo, altamente conservado en células eucariotas.
